氧化苦参碱微球冻干针剂器官靶向性研究

目的:研究氧化苦参碱微球(OMAT-PBCA-NP)冻干针剂的动物器官靶向性。方法:采用高效液相色谱法测定给药大鼠各主要脏器的氧化苦参碱含量。色谱条件:Kromasil C18柱(4.60 mm×250 mm,5μm),流动相甲醇-水-三乙胺(50∶50∶0.02),柱温20℃,检测波长210 nm,流速1 mL.min-1,进样量为10μL。结果:氧化苦参碱在50~400 mg.L-1线性关系良好(r=0.999 0),在30 min,1,4,8 h等时间点中,氧化苦参碱在肝内的含量最高,与其他脏器含量有极显著性差异(P<0.01)。结论OMAT-PBCA-NP具有良好的肝脏靶向性。 OBJECTIVE: To study the animal organ targeting of oxymatrine microspheres (OMAT-PBCA-NP) freeze-dried injection. Methods: The contents of oxymatrine in the main organs of rats were determined by high performance liquid chromatography. Chromatographic conditions: Kromasil C18 column (4.60 mm × 250 mm, 5 μm), mobile phase methanol-water-triethylamine (50:50:0.02), column temperature 20 ℃, detection wavelength 210 nm, , The injection volume of 10μL. Results: Oxymatrine showed a good linearity (r = 0.999 0) at 50-400 mg · L -1, and the highest content of oxymatrine in the liver at 30 min, 1, 4, 8 h (P <0.01), which was significantly different from other organs (P <0.01). Conclusion OMAT-PBCA-NP has good liver targeting.
