目的研究青蒿琥酯逆转人单核细胞白血病SHI-1细胞株多药耐药的效果。方法四甲基偶氮唑盐(MTT)法检测SHI-1细胞株对柔红霉素(DNR)、去甲柔红霉素(ID)、阿糖胞苷(Ara-c)、柔红霉素联合阿糖胞苷(DA)、去甲柔红霉素联合阿糖胞苷(IDA)的敏感性及青蒿琥酯逆转耐药的效果。结果 DA、IDA方案与单用化疗药相比,药物敏感性有提高,且差异具有统计学意义(P<0.05),非细胞毒性青蒿琥酯联合应用与单用化疗药、DA、IDA方案IC50差异具有显著统计学意义(P<0.001),随青蒿琥酯浓度的增加,逆转倍数呈上升趋势。结论联合应用非细胞毒剂量青蒿琥酯逆转SHI-1细胞株多药耐药作用显著,明显优于DA、IDA方案。
Objective To study the effect of artesunate on reversing multidrug resistance in human monocytic leukemia SHI-1 cell line. Methods Methyl thiazolyl tetrazolium (MTT) method was used to detect the effect of SHI-1 cell line on the expression of daunorubicin (DNR), daunorubicin (ID), Ara-c, (DA), daunorubicin combined with cytarabine (IDA) and the effect of artesunate on reversing drug resistance. Results The sensitivity of DA and IDA regimen was significantly higher than that of chemotherapy alone (P <0.05). The combination of non-cytotoxic artesunate with chemotherapy alone, DA and IDA IC50 difference was statistically significant (P <0.001), with the increase of artesunate concentration, the fold increase is on the rise. Conclusion The combination of non-cytotoxic doses of artesunate reverses SHI-1 cell line multidrug resistance was significantly better than the DA, IDA program.
