目的通过系统测定大鼠肝硬化发展过程中肝细胞凋亡的变化,与常规肝功能测定结果比较,了解上述指标与肝储备功能的关系。方法取Wistar大白鼠90只分为3组,分别为9周组、12周组、15周组,每组30只,采用复合因素(CCl4,乙醇)制备大鼠肝硬化模型;另取Wistar大白鼠30只随机分为3组,每组10只,作为与实验组相对应的对照组,代之等量生理盐水。分别于9周、12周、15周末处理9周组、12周组、15周组大鼠,同期处理相应对照组。血液标本作常规肝功检测;肝组织均作石蜡包埋,连续切片作HE染色、细胞凋亡检测。结果在大鼠肝硬化发展过程中HE染色显示9周组、12周组、15周组分别符合肝纤维化Ⅲ、Ⅳ期、肝硬化早期,肝细胞凋亡指数(AI)在9周组(7.38±1.71)、12周组(14.81±2.99)、15周组(26.55±7.94)时高于同期对照组(P<0.05)。结论①肝细胞凋亡指数随着肝硬化程度的加重而升高;②测定肝细胞凋亡指数对预测肝硬化患者的肝储备功能可能具有重要的意义。
Objective To determine the changes of hepatocyte apoptosis during the development of liver cirrhosis in rats and compare with the results of routine liver function test to understand the relationship between these indexes and liver reserve function. Methods Ninety Wistar rats were divided into three groups: 9 weeks, 12 weeks and 15 weeks, with 30 rats in each group. Rat model of liver cirrhosis was established by compounding factors (CCl4, ethanol) Thirty mice were randomly divided into 3 groups, 10 in each group, as the control group corresponding to the experimental group, instead of the same amount of normal saline. The rats in 9 weeks, 12 weeks and 15 weeks were treated with 9 weeks, 12 weeks and 15 weeks respectively, and the corresponding control group was treated at the same time. Blood samples for routine liver function tests; liver tissue were paraffin-embedded, serial sections for HE staining, apoptosis detection. Results Hematoxylin-eosin staining showed that in the development of hepatic cirrhosis of rats, HE staining showed that in the 9th, 12th and 15th week groups, they were respectively matched with stage Ⅲ and Ⅳ of liver fibrosis, early stages of liver cirrhosis and apoptosis index (AI) 7.38 ± 1.71), 12 weeks (14.81 ± 2.99), 15 weeks (26.55 ± 7.94), respectively, compared with the control group (P <0.05). Conclusion ① The index of hepatocyte apoptosis increased with the degree of liver cirrhosis. ② The determination of hepatocyte apoptosis index may have important significance in predicting the liver reserve function of patients with cirrhosis.
