目的筛选富集肉苁蓉中苯乙醇苷的最佳树脂和工艺条件,使分离纯化工艺达到最优化。方法选择7种大孔吸附树脂(AB-8、DM301、DM130、D1400、DA-201、HPD-100、D-101-I),通过树脂对苯乙醇苷的静态吸附和解析能力,筛选出最佳树脂,再通过树脂的静态特性和动态特性的筛选优化工艺。结果 DA-201的吸附解析最佳条件为:上样流速为1.5 mL.min-1,洗脱剂为90%的乙醇。按最佳工艺苯乙醇苷的纯度由25.1%上升为51.2%。结论该法简便、可行,为肉苁蓉中苯乙醇苷的纯化提供了参考。
Objective To screen and optimize the optimal resin and process conditions for the enrichment of phenylethanoid glycosides in Cistanche deserticola so as to optimize the separation and purification process. Methods Seven kinds of macroporous adsorption resins (AB-8, DM301, DM130, D1400, DA-201, HPD-100 and D-101-I) Good resin, and then through the static characteristics of resin and dynamic characteristics of the screening optimization process. Results The optimum conditions for the adsorption analysis of DA-201 were: the sample flow rate was 1.5 mL · min-1 and the eluent was 90% ethanol. The purity of phenylethanoid glycosides increased from 25.1% to 51.2% according to the best process. Conclusion The method is simple and feasible, which provides a reference for the purification of phenylethanoid glycosides in Cistanche tuberculosis.
