目的研究睾丸特异性基因BC051628在小鼠中的表达特征。方法通过分析小鼠基因表达谱芯片,发现BC051628基因特异性表达于睾丸组织。采用逆转录聚合酶链反应(RT-PCR)、实时聚合酶链反应(Real-time PCR)、蛋白免疫印迹法(Western blot)及免疫组化等方法分析该基因在小鼠不同组织及睾丸不同发育阶段的表达特征。利用生物信息学软件对该基因进行相关生物信息学分析。结果 BC051628基因在小鼠各组织中呈现睾丸特异性表达并伴随明显年龄依耐性。免疫组化染色发现BC051628蛋白主要定位于精原细胞、精母细胞及圆形精子。生物信息学分析发现BC051628基因编码蛋白氨基酸序列在人和小鼠具有高度同源性,提示该基因在哺乳动物进化过程中十分保守。结论 BC051628属于睾丸特异性基因,可能在生精过程发挥重要作用。
Objective To study the expression of testicular specific gene BC051628 in mice. Methods By analyzing mouse gene expression profiling chips, it was found that BC051628 gene was specifically expressed in testicular tissue. The gene was analyzed by RT-PCR, Real-time PCR, Western blot and immunohistochemistry in different tissues and testes of mice Developmental stages of the expression characteristics. The bioinformatics software was used to analyze the gene. Results The BC051628 gene showed testicular-specific expression in all tissues of mice with obvious age-dependent patience. Immunohistochemical staining found that BC051628 protein mainly located in spermatogonia, spermatocytes and round sperm. Bioinformatics analysis showed that BC051628 gene encoded protein amino acid sequence in human and mouse has a high degree of homology, suggesting that the gene is very conservative mammalian evolution. Conclusion BC051628 belongs to the testis-specific gene and may play an important role in the process of spermatogenesis.
