目的研究赤芍水提物对肝星状细胞株HSC-T6中bax、bcl-2、caspase-3蛋白表达的影响。方法采用乙醛造模后的肝星状细胞株HSC-T6细胞作为酒精性肝纤维化的体外研究模型;以赤芍水提物给彼格犬一次性灌胃,取给药后2h的血清作为实验药物血清;免疫细胞化学法检测肝星状细胞HSC-T6中bax、bcl-2、caspase-3蛋白表达。结果乙醛造模后的HSC-T6细胞中bcl-2的表达较正常组细胞增加,bax、caspase-3表达降低;药物血清作用后的肝星状细胞HSC-T6细胞中bcl-2的表达较模型组的HSC-T6细胞显著性降低,bax、caspase-3的表达较模型组细胞显著性增加。结论赤芍水提物可能是通过影响bax、bcl-2、caspase-3等基因异常表达来实现对HSC-T6的抑制增殖及促凋亡作用的,这可能与一些凋亡相关疾病如酒精性肝纤维化的发生有关。
Objective To investigate the effect of Radix Paeoniae Rubra on the expression of bax, bcl-2 and caspase-3 proteins in hepatic stellate cell line HSC-T6. METHODS: Hepatic stellate cell line HSC-T6 cells were used as an in vitro model for the study of alcoholic liver fibrosis. The mice were given a one-time intragastric administration of the red eel water extract and the serum was taken 2 h after administration. As the experimental drug serum; immunohistochemical detection of hepatic stellate cells HSC-T6 bax, bcl-2, caspase-3 protein expression. Results The expression of bcl-2 in HSC-T6 cells after acetaldehyde was increased compared with normal cells, and the expression of bax and caspase-3 was decreased. The expression of bcl-2 in hepatic stellate cells HSC-T6 cells after drug serum treatment Compared with the model group, the HSC-T6 cells were significantly decreased, and the expressions of bax and caspase-3 were significantly higher than those in the model group. Conclusion The water extract of Radix Paeoniae Rubra may be able to inhibit the proliferation and pro-apoptosis of HSC-T6 by affecting the abnormal expression of genes such as bax, bcl-2, caspase-3, which may be related to some apoptosis-related diseases such as alcoholicity. The occurrence of liver fibrosis is related.
