目的 :建立一种敏感的方法来检测肺癌患者外周血循环癌细胞 ,并探讨其临床应用的意义。方法 :采用反转录聚合酶链反应 (RT- PCR)技术检测肺癌患者外周血 CK19- m RNA的表达情况。结果 :40例肺癌外周血标本中 16例 (4 0 % ) CK19- m RNA阳性 ,15例肺良性病变中 1例 (6 .6 % )阳性 ,5例正常人为阴性。 10例有转移的肺癌患者 ,其外周血 CK19- m RNA均为阳性 (10 0 % ) ,30例检测时尚无转移证据者 ,有 6例阳性 (2 0 % ) ,经过半定量分析 ,两组 CK19- m RNA在量上差别有显著性。结论 :CK19- m RNA可作为一个标志物来检测肺癌患者外周血微转移 ;定量检测外周血 CK19- m RNA有助于诊断肺癌的血行转移 ;定期检测外周血 CK19- m RNA,尚可作为疗效的观察指标
Objective: To establish a sensitive method to detect circulating cancer cells in peripheral blood of patients with lung cancer and to explore its clinical significance. Methods: Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of CK19-m RNA in peripheral blood of patients with lung cancer. Results: 16 cases (40%) CK19-m RNA were positive in peripheral blood samples of 40 cases of lung cancer, 1 case (6. 6%) was positive in 15 cases of lung benign disease and 5 cases were negative in normal people. CK19-m RNA was positive in peripheral blood (10 0%) in 10 patients with metastatic lung cancer, 30 patients with no evidence of metastasis and 6 patients positive (20%). After semi-quantitative analysis, two groups CK19-m RNA differences in the amount of significant. Conclusion: CK19-m RNA can be used as a marker to detect peripheral blood micrometastasis in lung cancer patients. Quantitative detection of CK19-m RNA in peripheral blood can help diagnose the hematogenous metastasis of lung cancer. Regular detection of CK19- The observation index
