目的应用高脂高糖饮食联合链脲佐菌素(STZ)构建巴马小型猪2型糖尿病(T2DM)模型,并探讨T2DM小型猪胰岛素抵抗(IR)与同型半胱氨酸(homocysteine,Hcy)的关系。方法选用健康雄性巴马小型猪10只,随机分为两组:对照组(n=5),喂养普通饲料;糖尿病模型组(n=5),喂养高脂高糖饲料,上述两组共喂养10个月。11个月初糖尿病模型组腹腔注射STZ 100 mg/kg,1周后重复1次;对照组腹腔注射等剂量柠檬酸-柠檬酸钠缓冲液。观察两组小型猪体重、空腹血糖(FPG)、空腹胰岛素(FINS)及血浆Hcy水平,同时计算胰岛素抵抗指数(HOMA-IR)。结果高脂高糖饲料喂养10个月后,模型组小型猪体重、FPG、FINS及HOMA-IR高于对照组(P<0.01);与对照组相比STZ给药后(12个月末实验结束时),模型组的FPG进一步升高,FINS水平明显下降(P<0.01)。模型组Hcy高于对照组(P<0.01),且HOMA-IR与Hcy呈正相关(r=0.906,P<0.05)。结论T2DM小型猪Hcy升高与IR密切相关。
OBJECTIVE: To establish a T2DM model of Parmesan mini-pig with high-fat and high-sugar diet combined with streptozotocin (STZ) and investigate the relationship between insulin resistance and homocysteine (Hcy) Relationship. Methods Ten healthy male Bama miniature pigs were randomly divided into two groups: control group (n = 5) fed normal diet, diabetic model group (n = 5) fed high fat and high glucose diet, the above two groups were fed 10 months. At the beginning of 11 months, STZ was injected into the diabetic model group at a dose of 100 mg / kg. The control group was injected intraperitoneally with an equal dose of citric acid-sodium citrate buffer solution. The body weight, fasting blood glucose (FPG), fasting insulin (FINS) and plasma Hcy in the two groups of pigs were observed, and the insulin resistance index (HOMA-IR) was calculated. Results Compared with the control group, body weight, FPG, FINS and HOMA-IR of the model group were significantly increased (P <0.01) after 10 months of feeding with high-fat and high-sugar diet ), The FPG in model group increased further, and the level of FINS decreased significantly (P <0.01). Hcy in model group was higher than that in control group (P <0.01), and HOMA-IR was positively correlated with Hcy (r = 0.906, P <0.05). Conclusions The elevated Hcy in T2DM minipig is closely related to IR.
