目的:探讨大鼠β细胞素在胰腺干细胞的增殖和转分化为胰岛细胞中的生物学作用。方法:分离并培养大鼠胰腺干细胞于CMRL1066和无血清DMEM/F12培养液中,不同浓度大鼠β细胞素处理后,双硫腙染色,放免法检测胰岛素分泌量。结果:在大鼠β细胞素浓度为20~30mg/L时,大鼠胰腺干细胞转分化为胰岛样细胞团的数量明显高于对照组;其胰岛素分泌量明显高于对照组(P<0.01)。结论:大鼠β细胞素具有促进胰腺干细胞转分化为胰岛β细胞并增强其胰岛素分泌的作用。
OBJECTIVE: To investigate the biological role of rat β-catenin in the proliferation and transdifferentiation of pancreatic stem cells into islet cells. Methods: Rat pancreatic stem cells were isolated and cultured in CMRL1066 and serum-free DMEM / F12 medium. After treated with different concentrations of β-cytokines, dithizone staining and radioimmunoassay were used to detect insulin secretion. Results: The number of pancreatic stem cells transdifferentiated to islet-like cells was significantly higher than that of the control group at the concentration of 20-30 mg / L. The insulin secretion was significantly higher than that of the control group (P <0.01) . Conclusion: β-Cytokines can promote the transdifferentiation of pancreatic stem cells into islet β-cells and enhance insulin secretion.
