作者建立了一种SD大鼠的过敏性哮喘炎症模型的定量分析方法,用卵白蛋白(OA)低剂量多点皮下注射法致敏SD大鼠,2周后气雾吸入1%OA15min,吸入后6h和24h可见支气管-肺泡灌洗液中的白细胞计数显著增加,其中嗜酸性粒细胞增加较明显;肺切片显示细支气管和小血管周围水肿、大量嗜酸性粒细胞渗出浸润;计算机图象分析表明,细支气管管壁面积及管壁单位面积内嗜酸性粒细胞数增加显著。结果表明,抗原可诱导SD大鼠气道过敏性炎症,可以作为研究过敏性支气管哮喘的动物模型。
The authors established a quantitative analysis of allergic asthma in SD rat model. SD rats were sensitized to low dose of ovalbumin (OA) by multiple subcutaneous injections. After 2 weeks, aerosol inhalation of 1% OA15min, after inhalation 6h and 24h visible bronchoalveolar lavage fluid leucocyte count increased significantly, including eosinophils increased more obvious; lung section showed bronchioles and small blood vessels around the edema, a large number of eosinophil infiltration infiltration; computer image analysis The results showed that the area of the bronchioles and the number of eosinophils in the unit area of the wall increased significantly. The results showed that the antigen can induce airway allergic inflammation in SD rats and could be used as an animal model to study allergic bronchial asthma.
