目的探讨丹参酮IIA对人肝癌细胞TGF-β1表达的影响及其抗肿瘤作用机制。方法体外培养SMMC-7721肝癌细胞株,采用倒置相差显微镜、Hoechst33342染色荧光显微镜观察,以及“梯状”DNA确定凋亡的发生并计算凋亡率;用RT-PCR方法检测肝癌细胞TGF-β1的表达。结果倒置相差显微镜观察可见各试验组细胞的生长不同程度受抑,并可见典型的凋亡形态改变;Hoechst33342染色可见凋亡细胞核浓染,亮度明显加深或有染色质边集现象,亦可见典型的凋亡小体;提取基因组DNA电泳检测,出现典型的梯状条带;RT-PCR检测显示,丹参酮IIA作用48小时后TGF-β1表达量随药物浓度的增大而逐渐减低。结论丹参酮IIA可抑制人肝癌细胞SMMC-7721的细胞增殖,促进其凋亡,此作用可能与细胞内TGF-β1表达下调有关。
Objective To investigate the effect of tanshinone IIA on the expression of TGF-β1 in human hepatoma cells and its anti-tumor mechanism. Methods SMMC-7721 hepatocellular carcinoma cell line was cultured in vitro. The expression of TGF-β1 in hepatocellular carcinoma cells was detected by inverted phase contrast microscope, Hoechst33342 staining fluorescence microscope and “ladder DNA” expression. Results Inverted phase contrast microscopy showed that the growth of cells in various experimental groups was inhibited to varying degrees, and typical morphological changes of apoptosis were observed. Hoechst33342 staining showed apoptotic nuclei staining with markedly enhanced brightness or chromatin edge-set phenomenon, as well as typical Apoptotic bodies; typical genomic DNA was detected by electrophoresis, and the typical ladder-like bands appeared. RT-PCR showed that the expression of TGF-β1 decreased gradually with the increase of drug concentration after tanshinone IIA treatment for 48 hours. Conclusion Tanshinone IIA can inhibit the proliferation and promote the apoptosis of human hepatocellular carcinoma cell line SMMC-7721, which may be related to the down-regulation of TGF-β1 expression.
