目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞PDCD-1的表达及其在SLE发病中的意义。方法用荧光标记的单克隆抗体及流式细胞仪术测定36例SLE和20例正常人外周血中性粒细胞、淋巴细胞PDCD-1的表达,并将其与抗核抗体(ANA)、抗ds-DNA抗体、SLEDAI积分做相关性分析。结果SLE患者外周血中性粒细胞、淋巴细胞PDCD-1的表达异常,中性粒细胞、淋巴细胞PDCD-1的表达较正常人降低(P<0.02),淋巴细胞PDCD-1表达与抗ds-DNA抗体、SLEDAI积分呈低度相关(r=0.343,r=0.335,P<0.05)。结论SLE患者外周血中性粒细胞、淋巴细胞PDCD-1的表达异常,不能维持机体的正常免疫耐受平衡,在SLE的免疫发病机制中起重要的作用。
Objective To investigate the expression of PDCD-1 in peripheral blood mononuclear cells in patients with systemic lupus erythematosus (SLE) and its significance in the pathogenesis of SLE. Methods The expression of PDCD-1 in peripheral blood mononuclear cells and lymphocytes of 36 patients with SLE and 20 normal controls were detected by fluorescence-labeled monoclonal antibody and flow cytometry. The expressions of PDCD-1 and anti-nuclear antibody (ANA) ds-DNA antibody, SLEDAI score correlation analysis. Results The expression of PDCD-1 in peripheral blood mononuclear cells and lymphocytes in patients with SLE was abnormal, the expression of PDCD-1 in neutrophils and lymphocytes was lower than that in normal subjects (P <0.02), and the expression of PDCD-1 in lymphocytes was positively correlated with anti-ds -DNA antibody, SLEDAI score was low correlated (r = 0.343, r = 0.335, P <0.05). Conclusion The abnormal expression of PDCD-1 in peripheral blood neutrophils and lymphocytes in SLE patients can not maintain the balance of normal immune tolerance in SLE patients and play an important role in the pathogenesis of SLE.
