目的检测核因子(NF-κB)亚基P65及抑制因子(ⅠκBα)在糖尿病肾病发病过程中肾组织中的表达,以探讨NF-κB、ⅠκBα在糖尿病肾病发病中的作用。方法2003-09~2004-11在中国医科大学第一医院以链脲菌素(STZ)制备糖尿病大鼠模型2周、4周、12周组;检测血肌酐和尿蛋白和用ABC法检测NF-κBP65、ⅠκBα在肾小球的表达。结果在糖尿病肾病发病过程中ⅠκBα胞浆表达的阳性细胞数逐渐减弱;NF-κBP65核内表达的阳性细胞数逐渐升高。ⅠκBα胞浆表达与糖尿病肾损害呈负相关;NF-κBP65核内表达与肾损害呈正相关;ⅠκBα胞浆表达与NF-κBP65核内表达呈负相关。结论NF-κB与ⅠκBα在糖尿病肾病发病过程中的肾小球的相反表达方式与糖尿病肾病的发生、发展密切相关,ⅠκBα发挥关键作用。
Objective To detect the expression of nuclear factor (NF-κB) subunit P65 and inhibitory factor (IκBα) in renal tissues during the pathogenesis of diabetic nephropathy, and to explore the role of NF-κB and IκBα in the pathogenesis of diabetic nephropathy. Methods From September 2003 to November 2004, diabetic rats were treated with streptozotocin (STZ) for 2 weeks, 4 weeks and 12 weeks in the First Hospital of China Medical University. Serum creatinine and urinary protein were detected by enzyme linked immunosorbent assay (ELISA) and NF -κBP65, IκBα expression in the glomerulus. Results The number of positive cells in IκBα cytoplasm gradually decreased during the pathogenesis of diabetic nephropathy. The number of positive cells in nucleus of NF-κBβ65 increased gradually. The expression of IκBα in cytoplasm was negatively correlated with diabetic nephropathy. The expression of NF-κBp65 in nucleus was positively correlated with renal damage. The expression of IκBα was negatively correlated with the expression of nuclear NF-κB p65. Conclusion The inverse expression of NF-κB and IκBα in glomeruli of diabetic nephropathy is closely related to the occurrence and development of diabetic nephropathy, and IκBα plays a key role.
