目的研究大鼠局灶性脑缺血过程中c_fosmRNA表达的动态变化 ,并观察小檗碱(Ber)对其作用。方法运用斑点杂交技术 ,观察Ber对局灶性脑缺血再灌注大鼠大脑皮质及海马组织c_fosmRNA表达的影响。结果大鼠局灶性脑缺血2h再灌注0.5~4h ,脑皮质及海马组织c_fosmRNA出现一过性高表达 ,2h达高峰 ,分别为对照组的4.5和4.7倍。Ber20mg/(kg·d)ip显著抑制大鼠脑缺血诱导的c_fos高表达 ,降低病灶侧海马和皮层组织水、钙含量。结论脑缺血过程中c_fos原癌基因呈现一过性高表达 ,Ber可降低c_fosmRNA水平 ,该作用可能是其抗脑缺血机理之一
Objective To study the dynamic changes of c_fos mRNA expression during focal cerebral ischemia in rats and observe the effect of berberine (Ber) on it. Methods Dot blotting was used to observe the effect of Ber on the expression of c_fos mRNA in cerebral cortex and hippocampus of rats after focal cerebral ischemia/reperfusion. Results After focal cerebral ischemia for 2h and reperfusion for 0.5-4h, c-fos mRNA in the cerebral cortex and hippocampus was transiently overexpressed and peaked at 2h, 4.5 and 4.7 times of the control group, respectively. Ber20mg/(kg·d)ip significantly inhibited the high expression of c_fos induced by cerebral ischemia in rats and decreased the content of water and calcium in hippocampus and cortex tissue of lesion side. Conclusion The c_fos proto-oncogene is transiently over-expressed during cerebral ischemia, and Ber may decrease the level of c_fos mRNA, which may be one of the mechanisms of anti-cerebral ischemia.
