目的测定人巨噬细胞金属弹性蛋白酶(HME)在胃癌细胞及胃癌组织中的表达,评价其在胃癌进展中的作用。方法 2003年4月至8月胃肠外科和肿瘤外科胃癌住院手术病人共58例,所有病人均取肿瘤组织、癌旁组织、远处正常组织标本,采用 RT-PCR、实时荧光定量 PCR 测定胃癌细胞株(MGC-803、SGC-7901、AGS)及胃癌组织中 HME mRNA 水平,采用 Western 印迹和免疫组化法测定 HME 蛋白水平。结果 3株胃癌细胞株中均有 HME mRNA 和蛋白表达;胃癌组织中的 HMEmRNA 和蛋白表达高于远处正常组织,差异有统计学意义(P<0.05);而胃癌组织与癌旁组织中的HME mRNA 和蛋白表达差异无统计学意义(P>0.05)。结论胃癌患者中 HME mRNA 和蛋白呈一定比例的阳性表达,胃癌组织中最高,其次为癌旁组织和远处正常组织,说明 HME 是一项有潜在价值的肿瘤相关标志物。
Objective To determine the expression of human macrophage metalloelastase (HME) in gastric cancer cells and gastric cancer and to evaluate its role in the progression of gastric cancer. Methods A total of 58 hospitalized patients with gastric cancer who underwent gastric surgery and gastric cancer from April to August 2003 were enrolled in this study. Tumor tissues, pericancerous tissues and distant normal tissues were collected from all patients. Gastric cancer was detected by RT-PCR and real-time fluorescence quantitative PCR The levels of HME mRNA in gastric cancer cell lines (MGC-803, SGC-7901, AGS) and gastric cancer tissues were determined by Western blotting and immunohistochemistry. Results The expression of HME mRNA and protein were found in all three gastric cancer cell lines. The expression of HME mRNA and protein in gastric cancer tissues was higher than that in distant normal tissues (P <0.05), while the expression of HME mRNA and protein in gastric cancer tissues was significantly higher than that in adjacent tissues HME mRNA and protein expression difference was not statistically significant (P> 0.05). Conclusion The positive expression of HME mRNA and protein in gastric cancer patients is the highest, followed by the paracancer tissues and distant normal tissues, indicating that HME is a potential tumor-associated marker.
