目的 :用 AP- PCR方法检测新生隐球菌的临床和环境分离株 ,观察 PCR指纹与血清型的关系 ,探索 PCR指纹对于新生隐球菌的分型标准。方法 :采用 3种寡核苷酸重复序列即 CN1 [(GTG) 5]、CN2 [(GACA) 4]和 CN3 [(GATA) 4]为引物对从患者和鸽粪中分离到的 48株新生隐球菌 DNA进行扩增 ,根据扩增产物的电泳带型来对比 PCR指纹和血清型之间的关系。结果 :38株血清 A型株中 ,34株有完全相似的 DNA带型 ,另 4株中 1株与血清 D型的 DNA带型相似 ,3株的 PCR指纹与血清 A、B、C、D型的带型均不相同 ,各有独特的 PCR指纹带型。 2株 B型和 2株 C型产生几乎完全一致的 PCR指纹带型 ,3株 D型和 3株其他隐球菌则各自产生特定的 DNA带型。 结论 :(1)大多数血清 A型新生隐球菌株具有相似 PCR指纹带型 ;(2 )血清 B型和 C型 (gattii变种 )的 PCR指纹不易区别 ,相同血清型不同的株可能有不同的 PCR指纹带型 ;(3) PCR指纹法用于新生隐球菌菌株鉴定是一种快速、方便、可行的方法
OBJECTIVE: To detect the clinical and environmental isolates of Cryptococcus neoformans by AP-PCR and to observe the relationship between PCR fingerprints and serotypes and explore the PCR fingerprinting criteria for C. neoformans. Methods: 48 neonates isolated from patients and pigeon feces were obtained using three kinds of oligonucleotide repeats, CN1 [(GTG) 5], CN2 [(GACA) 4] and CN3 [(GATA) 4] Cryptococcus DNA amplification, amplification products according to the electrophoresis banding pattern to compare the relationship between PCR fingerprints and serotypes. Results: Of the 38 serotype A strains, 34 had similar DNA bands, and the other 4 belonged to serotype D DNA bands. The PCR fingerprints of 3 strains were similar to those of serum A, B, C, D Types of tape are not the same, each with a unique PCR fingerprint band. Two strains of type B and two strains of type C produced almost identical PCR fingerprinting, and three strains of type D and three strains of other cryptococcosis each produced a specific DNA pattern. Conclusion: (1) Most strains of Cryptococcus neoformans serotype A have similar PCR fingerprinting patterns. (2) PCR fingerprints of serotypes B and C (gattii variant) are not easy to distinguish, and strains with the same serotype may have different PCR fingerprinting PCR fingerprinting; (3) PCR fingerprinting method for the identification of C. neoformans strain is a fast, convenient and feasible method
