目的观察维生素E(VE)及镁联合对糖尿病大鼠胰岛素抵抗和抗氧化水平的影响。方法88只大鼠随机分为6组,分别喂饲含有不同剂量VE及镁的饲料4周;大鼠腹腔注射链脲咗菌素诱导糖尿病腜?继续喂养4周处死动物,计算胰岛素抵抗指数;测定谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)水平;检测红细胞溶血度。结果与对照组比较,VE、VE+中、高剂量镁组胰岛素抵抗指数分别降低了5.52,3.29和2.65(P<0.05);VE+高剂量镁组GSH-Px与SOD分别为(41.65±2.99)和(119.49±8.03)U/(mg.prot),明显高于对照组(P<0.05);VE组、中、高剂量镁组、VE+中、高剂量镁组红细胞溶血度分别降低了67.86%,71.43%,60.71%,89.29%和89.29%(P<0.05)。结论联合补充VE和镁可以通过提高机体抗氧化水平降低胰岛素抵抗,较单纯补充VE或镁效果更佳。
Objective To observe the effects of vitamin E (VE) and magnesium on the insulin resistance and antioxidation in diabetic rats. Methods Eighty-eight rats were randomly divided into 6 groups and were fed diets containing different doses of VE and magnesium for 4 weeks respectively. Rats were injected with streptozotocin to induce diabetes mellitus. Animals were sacrificed after 4 weeks and insulin resistance index was calculated. Determination of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) levels; detection of hemolysis of erythrocytes. Results Compared with the control group, the insulin resistance index decreased by 5.52, 3.29 and 2.65 respectively (P <0.05) in VE, VE + medium and high dose magnesium groups; GSH-Px and SOD in VE + high dose magnesium group were (41.65 ± 2.99) (119.49 ± 8.03) U / (mg.prot), which was significantly higher than that of the control group (P <0.05). The hemolysis of erythrocytes in VE group, medium and high dose magnesium group and medium and high dose magnesium group decreased 67.86% 71.43%, 60.71%, 89.29% and 89.29%, respectively (P <0.05). Conclusions Combination of VE and magnesium can reduce insulin resistance by increasing body antioxidant level, which is more effective than simply supplementing VE or magnesium.
