目的探讨环氧合酶-2(COX-2)、基质金属蛋白酶-2(MMP-2)在乳腺浸润性癌中的表达情况及它们之间的相互关系。方法采用免疫组化SP法检测100例乳腺癌及40例正常乳腺组织中COX-2、MMP-2的表达情况。结果 COX-2蛋白在正常乳腺组织及乳腺癌中的阳性表达率分别为7.5%(3/40)、83.0%(83/100),而且COX-2在淋巴结转移组中的阳性表达率[90.0%(54/60)]明显高于无淋巴结转移组(P<0.05)。MMP-2蛋白在正常乳腺组织及乳腺癌中的阳性表达率分别为0(0/40)、94.0%(94/100),而且MMP-2在淋巴结转移组中的阳性表达率[98.3%(59/60)]明显高于无淋巴结转移组[87.5%(35/40)](P<0.05)。COX-2和MMP-2表达呈正相关关系(r=0.334,P<0.05)。结论检测COX-2、MMP-2表达将有助于判断乳腺癌的转移潜能及预后。
Objective To investigate the expression of cyclooxygenase-2 (COX-2) and matrix metalloproteinase-2 (MMP-2) in breast invasive carcinoma and their relationship. Methods The expression of COX-2 and MMP-2 in 100 cases of breast cancer and 40 cases of normal breast tissue were detected by immunohistochemical SP method. Results The positive expression rates of COX-2 protein in normal breast tissues and breast cancer tissues were 7.5% (3/40) and 83.0% (83/100), respectively. The positive expression rates of COX-2 in lymph node metastasis group were 90.0% % (54/60)] was significantly higher than those without lymph node metastasis (P <0.05). The positive rates of MMP-2 protein in normal breast tissues and breast cancer tissues were 0 (0/40) and 94.0% (94/100) respectively, and the positive rate of MMP-2 in lymph node metastasis was 98.3% ( 59/60)] was significantly higher than those without lymph node metastasis [87.5% (35/40)] (P <0.05). There was a positive correlation between the expression of COX-2 and MMP-2 (r = 0.334, P <0.05). Conclusion Detecting the expression of COX-2 and MMP-2 will help to determine the metastatic potential and prognosis of breast cancer.
