目的 研究有关生化因素对喜树碱(CPT)诱导人鼻咽癌细胞凋亡的影响。方法 在CPT诱导人鼻咽癌CNE-2Z 细胞凋亡的基础上,采用流式细胞仪及二苯胺反应等方法,观察了多胺、Ca2+、Mg2十依赖性内切酶活性抑制剂及PARP活性抑 制剂对DNA片段化形成的影响。结果 多胺可明显抑制CPT诱导的CNE-2Z细胞凋亡,使片段化DNA明显降低,P<0.05。内 切酶及PARP活性抑制剂与CPT共同作用CNE-2Z细胞后测得片段化DNA与单用CPT相比无显著性差异,P>0.05。结论 喜树碱诱导的 CNE-2Z细胞凋亡可能与一种非 Ca2+、Mg2+依赖性内切酶活性有关,且受多胺的调节。
Objective To investigate the effects of biochemical factors on the apoptosis of human nasopharyngeal carcinoma cells induced by CPT. Methods CPT induced CNE-2Z cell apoptosis in human nasopharyngeal carcinoma based on the use of flow cytometry and diphenylamine reaction and other methods were observed Polyamine, Ca2, Mg2-dependent endonuclease activity inhibitor and PARP activity Effect of inhibitors on DNA fragmentation. Results Polyamines significantly inhibited the apoptosis of CNE-2Z cells induced by CPT and markedly reduced the fragmented DNA (P <0.05). Endonuclease and PARP inhibitor combined with CPT in CNE-2Z cells measured fragmented DNA and CPT alone was no significant difference, P> 0.05. CONCLUSION Camptothecin-induced apoptosis of CNE-2Z cells may be related to a non-Ca2 +, Mg2 + -dependent endonuclease activity and is regulated by polyamines.
