血清甲状腺素(Tetfaiodothyronine;Thyro xine,简称T_4)RIA含量测定是目前临床上常用于诊断、鉴别诊断“甲亢”或“甲减”症,甲亢症及甲状腺肿瘤病人手术前的准备、治疗效果观察、随访复查、研究探讨甲状腺疾病以及甲状腺激素与其他疾病关系的指标之一,是对甲状腺功能判断最基本的筛选试验。 一、T_4放射免疫测定的基本原理:已知适当浓度的T_4特异性抗体与血清T_4和I-T_4之间竞争地结合,结合到抗体上的I—T_4部分与未标记T_4的浓度呈反比关系,利用分离剂将分离结合标记抗原和游离标记抗原,测量沉淀部分的放射性,根据结合率标准曲线,样品中T_4浓度就能查知。 二、参考正常值:上海放射免疫分析技术研究所生产的血清T_4 RIA试剂盒,正常值为45~130ng/ml。根据文献报道,不同年龄、地区及实验室,参考正常值有如下几种情况。 (一)新生儿出生后三天内的血清:1天18.6±4.39μg%(范围11~28μg%);2天15.0±2.80μg%(范囱9~20μg%);3天14.9±4.33μ%(范围8.4~24μg%)。
