目的 探讨肝结散对纤维化小鼠血清和肝脏组织匀浆中Ⅲ型前胶原 (procollagentype Ⅲ ,PCⅢ )、透明质酸 (hyaluronicacid ,HA)及层粘连蛋白 (laminin ,LN)等三种ECM成分含量的影响。方法 以肝结散、秋水仙碱抗肝纤维化治疗四氯化碳 (CCl4)诱导的肝纤维化模型小鼠 ,并分别在治疗开始后的第 4、8、12周 ,以常规方法观察肝功能和肝纤维化程度的变化 ,以放射免疫分析法测定其血清和肝脏组织匀浆中PCⅢ、HA、LN等三种ECM成分的含量。结果 模型组小鼠上述指标的含量在第 4周开始升高 ,且在整个观察期内随肝功能损害加重和肝纤维化程度加深而逐渐增加。以肝结散治疗后 ,小鼠肝功能损害减轻 ,纤维化程度降低 ,上述指标在血清和肝组织匀浆中的含量减少 (P <0 0 1)。结论 肝结散对以PCⅢ、HA、LN为代表的肝细胞外基质 (ECM)的沉积与分解代谢可能有直接的影响
Objective To investigate the effect of liver distention on the three kinds of ECM components including procollagen type III (PCIII), hyaluronic acid (HA) and laminin (LN) in serum and liver homogenate of mice with fibrosis. The effect of content. Methods Hepatic fibrosis and colchicine were used to treat hepatic fibrosis induced by carbon tetrachloride (CCl4) in mice. The liver was observed by routine methods at the 4th, 8th, and 12th weeks after the start of treatment. Changes in function and liver fibrosis were determined by radioimmunoassay to determine the contents of PCIII, HA, and LN in the serum and liver homogenates. Results The content of above-mentioned indicators in the model group began to increase at the 4th week, and gradually increased with the worsening of liver function damage and the deepening of liver fibrosis during the whole observation period. After treatment with hepatic disintegration, the hepatic function of the mice was reduced and the degree of fibrosis was reduced. The above-mentioned indexes were reduced in serum and liver homogenates (P < 0 01). Conclusion Hepatic deposition may directly affect the deposition and catabolism of hepatic extracellular matrix (ECM) represented by PCIII, HA, and LN.
