目的:观察Dlx1/2在经Kcl刺激大鼠E 17脑皮质神经细胞中的表达,揭示该对基因在核酸与蛋白水平的表达特点及其在大脑接受刺激后反应早期功能活动中的初步作用。方法:原代细胞培养获取SD大鼠E17脑皮质神经细胞;qRT-PCR分析2个基因的核酸在Kcl刺激0~24 h中的表达变化情况;Western blot分析2个基因的蛋白在Kcl刺激0~24 h中的表达变化情况。结果:原代细胞培养显示细胞状态良好。qRT-PCR显示Dlx1/2在经Kcl刺激细胞的24 h内mRNA表达趋势均为先上升后下降,其中6 h是高峰,24 h出现下降。Western blot显示经Kcl刺激后,Dlx1的表达增高只出现在12 h内,24 h出现明显下降;而Dlx2表达随时间延长而明显增高,24 h稍有下降。结论:Dlx1/2的表达与Kcl刺激时间的长短有关,Dlx2表达整体高于Dlx1。Dlx1/2表达升高对胚胎期脑皮质神经细胞早期反应中的活动功能具有重要作用,其中Dlx2的作用可能相对更大。
OBJECTIVE: To observe the expression of Dlx1 / 2 in neurons of E 17 -stimulated rat induced by KCl, and to reveal its expression at nucleic acid and protein levels and its preliminary role in early functional reaction after stimulation of brain. Methods: The primary cultured rat E17 cortical neurons were obtained by primary cell culture. The expression of two genes was detected by qRT-PCR at 0-24 h after Kcl stimulation. ~ 24 h in the expression changes. Results: Primary cell culture showed good cell state. The results of qRT-PCR showed that the expression of Dlx1 / 2 in 24 h after KCl stimulation increased first and then decreased, and peaked at 6 h and decreased at 24 h. Western blot showed that the expression of Dlx1 increased only within 12 h after Kcl stimulation, but decreased significantly at 24 h. However, the expression of Dlx2 increased significantly with time and decreased slightly at 24 h. Conclusion: The expression of Dlx1 / 2 is related to the duration of Kcl stimulation and the overall expression of Dlx2 is higher than that of Dlx1. The increased expression of Dlx1 / 2 plays an important role in the activation of embryonic cortical neurons in early response, and the effect of Dlx2 may be relatively greater.
