目的观察槲皮苷防治溃疡性结肠炎及抗炎、镇痛、止泻等药效。方法以醋酸灌肠复制溃疡性结肠炎模型,大鼠从造模前2d至造模后6dig给予槲皮苷(50,200mg·kg-1·d-1),取结肠粘膜进行组织病理学检查以及丙二醛(MDA)、髓过氧化物酶(MPO)测定。小鼠ig给予槲皮苷(75,300mg·kg-1·d-1)4d后测定腹腔毛细血管通透性、醋酸ip引起扭体反应以及番泻叶煎剂ig引起腹泻潜伏期及次数变化。结果槲皮苷减轻溃疡性结肠炎大鼠结肠病变程度,降低结肠粘膜组织MDA及MPO水平(P<0.05);抑制醋酸引起的小鼠腹腔毛细血管通透性增加,减少扭体次数(P<0.05);对腹泻潜伏期及腹泻次数无显著影响(P>0.05)。结论槲皮苷对大鼠溃疡性结肠炎具有防治作用,并有抗炎、镇痛作用。
Objective To observe the effect of quercetin in the treatment of ulcerative colitis and anti-inflammatory, analgesic and antidiarrheal effects. Methods The model of ulcerative colitis was duplicated by acetic acid enema. Rats were given quercitrin (50,200 mg·kg-1·d-1) from 2 days before modeling to 6 days after modeling. Colonic mucosa was taken for histopathological examination and C Dialdehyde (MDA), myeloperoxidase (MPO) assay. After ig administration of quercitrin (75,300 mg·kg-1·d-1) to mice for 4 days, the capillary permeability of the abdominal cavity, writhing response induced by acetic acid ip, and diarrhea latency caused by sigma decoction ig were determined. RESULTS Quercitrin alleviated the degree of colonic lesions in rats with ulcerative colitis, decreased the MDA and MPO levels in the colonic mucosal tissue (P<0.05); inhibited acetic acid-induced increases in capillary permeability of the abdominal cavity in mice, and reduced writhing frequency (P< 0.05); There was no significant effect on the latent period of diarrhea and the number of diarrhea (P>0.05). Conclusion Quercitrin has preventive and therapeutic effects on ulcerative colitis in rats, and has anti-inflammatory and analgesic effects.
