目的研究三磷酸腺苷结合盒转运蛋白G2基因(adenosine triphosphate-binding cassette transporter protein G2 gene,ABCG2)在原发性痛风性关节炎(gouty arthritis,GA)患者中检测及临床意义。方法收集2015-08/2016-08月GA患者60例为实验组,根据发病时间长短分为原发性急性痛风性关节炎(acute gouty arthritis,AGA)和原发性间歇性急性痛风性关节炎(intermittent gouty arthritis,IGA)两组各30例,选择同期健康体检者20例为对照组,采用聚合酶链反应(polymerase chain reaction,PCR)测所有标本外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)中ABCG2 mRNA表达,同时测定尿酸(uric acid,UA)、尿素氮(urea nitrogen,UN)、甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)等指标。结果实验组中ABCG2 mRNA水平高于对照组,组间比较差异具有统计学意义(P<0.05);AGA组和IGA组ABCG2 mRNA水平均高于对照组,组间比较差异具有统计学意义(P<0.05);痛风患者中ABCG2 mRNA水平与UA水平呈正相关(r=0.28,P<0.05),而与其他生化指标无明显相关性。结论 ABCG2基因在痛风患者中有高水平表达,该基因的检测对阐明GA发病机制提供理论依据。
Objective To study the detection and clinical significance of adenosine triphosphate-binding cassette transporter protein G2 gene (ABCG2) in patients with primary gouty arthritis (GA). Methods Sixty patients with GA in our hospital from January 2015 to August 2016 were enrolled in this study. The patients were divided into primary acute gouty arthritis (AGA) and primary intermittent acute gouty arthritis (30 cases) with intermittent gouty arthritis (IGA). 20 healthy subjects were selected as the control group. All the peripheral blood mononuclear cells (PBMCs) were detected by polymerase chain reaction (PCR) (ABCG2) mRNA expression in PBMCs were measured. Uric acid (UA), urea nitrogen (UN), triglyceride (TG) and total cholesterol (TC) were also measured. Results The level of ABCG2 mRNA in the experimental group was significantly higher than that in the control group (P <0.05). The levels of ABCG2 mRNA in AGA group and IGA group were significantly higher than those in the control group (P <0.05). There was a positive correlation between ABCG2 mRNA level and UA level in gout patients (r = 0.28, P <0.05), but not with other biochemical parameters. Conclusion The ABCG2 gene is highly expressed in gout patients. The detection of this gene provides a theoretical basis for elucidating the pathogenesis of.
