目的探讨脑出血(ICH)后血肿周围组织细胞黏附分子(ICAM-1)含量的变化及早期姜黄素治疗对其表达的影响。方法采用未抗凝新鲜自体股动脉血注入大鼠尾状核建立脑出血动物模型。将60只大鼠随机分成对照组、ICH组、姜黄素低干预组,于建模后给予300mg/(kg.d)姜黄素,连续腹腔注射7天,各组分6小时、1天、3天和7天共4个时间点。分别采用干重湿重法、免疫组织化学法测定不同时间点的脑组织水含量和ICAM-1表达的变化。结果脑组织水含量和ICAM-1表达均在ICH后6小时开始升高,3天达到高峰,7天下降至正常水平,两者之间存在明显的相关性(r=0.88,P<0.01)。姜黄素小剂量及中等剂量治疗能够降低各个时间点的脑组织水含量和ICAM-1的表达。结论 ICAM-1与脑水肿的形成密切相关;姜黄素治疗可以降低ICH后血肿周围脑组织ICAM-1的表达,减轻ICH后的脑水肿形成。
Objective To investigate the changes of intracellular ICAM-1 in peripheral blood after intracerebral hemorrhage (ICH) and the effect of early curcumin treatment on its expression. Methods The animal model of intracerebral hemorrhage was established by injecting the fresh autologous femoral artery without cautery into rat caudate nucleus. Sixty rats were randomly divided into control group, ICH group and low curcumin intervention group. Curcumin (300mg / (kg.d)) was given after modeling for 7 days. Each group was given 6 hours, 1 day, 3 days A total of 4 days and seven days. The changes of water content and ICAM-1 expression in brain tissue at different time points were measured by dry weight wet weight method and immunohistochemical method respectively. Results The brain water content and ICAM-1 expression increased 6 hours after ICH, reached the peak at 3 days and decreased to normal level at 7 days. There was a significant correlation between them (r = 0.88, P <0.01) . Curcumin treatment of low and medium dose can reduce the brain water content and ICAM-1 expression at various time points. Conclusion ICAM-1 is closely related to the formation of cerebral edema. Curcumin treatment can reduce the expression of ICAM-1 in the perihematomal brain tissue after ICH and reduce the formation of cerebral edema after ICH.
