目的:分析冬虫夏草及其功能食品中的主要活性成分。方法:选择不同溶剂提取冬虫夏草样品,比较它们的提取效果;采用高效液相色谱-电喷雾质谱结合选择离子监测(SIR)法分析冬虫夏草中虫草酸、虫草素、尿嘧啶等活性成分。结果:确立了样品提取的最佳溶剂;经色谱、质谱条件的优化,建立了定量分析冬虫夏草及其功能食品中活性成分的方法。虫草酸、尿嘧啶、次黄嘌呤、尿苷、腺嘌呤、腺苷、虫草素的定性离子分别为183、111、137、265、136、268、252,检出限(LOD)分别为74.2、72.0、85.0、82.0、4.2、50.0、65.0ng/mL(进样量5μL),回收率90%~110%。结论:该方法灵敏度高、选择性好、操作简便、定量准确,适用于冬虫夏草及其功能食品中的活性物质分析及产品质量控制。
Objective: To analyze the main active ingredients in Cordyceps sinensis and its functional foods. Methods: Different solvents were used to extract Cordyceps sinensis samples and their extraction effects were compared. High performance liquid chromatography-electrospray ionization mass spectrometry combined with selective ion monitoring (SIR) was used to analyze Cordyceps sinensis, cordycepin, uracil and other active ingredients. Results: The best solvent for sample extraction was established. After optimization of chromatographic and mass spectrometry conditions, a method for quantitative analysis of active ingredients in Cordyceps sinensis and its functional foods was established. The qualitative ions of cordycepic acid, uracil, hypoxanthine, uridine, adenine, adenosine, cordycepin were 183, 111, 137, 265, 136, 268, 252, respectively, and the limit of detection (LOD) was 74.2, respectively. 72.0, 85.0, 82.0, 4.2, 50.0, 65.0 ng/mL (injection volume 5μL), recovery rate of 90% to 110%. Conclusion: The method has high sensitivity, good selectivity, simple operation and accurate quantitative. It is suitable for the analysis of active substances in Cordyceps sinensis and its functional foods and product quality control.
