背景有研究证实,白介素6受体(IL-6R)基因中的rs4129267位点多态性与哮喘易患性显著相关,但尚无评估rs4129267位点多态性在汉族儿童中分布情况及其与血清IgE水平相关性的研究。目的研究IL-6R基因rs4129267位点多态性与汉族儿童哮喘易患性及血清IgE水平的关系。方法选取2013—2014年于广东省妇幼保健院儿科就诊的哮喘患儿410例为哮喘组,同期选取广东省妇幼保健院住院的非哮喘患儿513例为对照组。提取两组患儿基因组DNA,采用SNa Pshot法对IL-6R基因rs4129267、rs527236128位点进行基因分型,采用酶联免疫吸附试验(ELISA)法检测两组患儿血清总IgE水平。结果两组患儿rs4129267位点和rs527236128位点的基因型分布均符合Hardy-Weinberg平衡(χ~2=3.264、2.135,P>0.05)。两组患儿rs4129267位点基因型频率和等位基因频率比较,差异有统计学意义(P<0.05)。两组患儿rs527236128位点基因型频率和等位基因频率比较,差异无统计学意义(P>0.05)。Logistic回归分析结果显示,TT基因型是儿童哮喘的影响因素(P<0.05)。对照组患儿血清总IgE水平低于哮喘组(P<0.05)。对照组、哮喘组组内不同基因型患儿血清总IgE水平比较,差异无统计学意义(P>0.05);对照组各基因型患儿血清总IgE水平均低于哮喘组(P<0.05)。结论 IL-6R基因rs4129267位点多态性与汉族儿童哮喘易患性有关,而与哮喘患儿血清IgE水平无关。
Background Studies have shown that rs4129267 polymorphism in interleukin 6 receptor (IL-6R) gene is significantly associated with asthma susceptibility, but no distribution of rs4129267 polymorphism in Han children and its association with Study on the Correlation of Serum IgE Levels. Objective To investigate the association of rs4129267 polymorphism of IL-6R gene with asthma susceptibility and serum IgE levels in Han children. Methods A total of 410 children with asthma were selected from pediatric department of Guangdong MCH in 2013-2014 as asthma group. 513 non-asthmatic children hospitalized in Guangdong MCH hospital were selected as the control group. Genomic DNA was extracted from both groups. Genotypes of rs4129267 and rs527236128 of IL-6R gene were genotyped by SNa Pshot method. Serum total IgE levels were measured by enzyme-linked immunosorbent assay (ELISA). Results The genotypes of rs4129267 and rs527236128 in both groups were in accordance with Hardy-Weinberg equilibrium (χ ~ 2 = 3.264, 2.135, P> 0.05). There were significant differences in genotype frequency and allele frequency between rs4129267 loci in two groups (P <0.05). There was no significant difference in genotype frequency and allele frequency between rs527236128 loci in two groups (P> 0.05). Logistic regression analysis showed that TT genotype was the influencing factor of childhood asthma (P <0.05). The serum total IgE level in control group was lower than that in asthma group (P <0.05). There was no significant difference in serum total IgE levels between the control group and the asthma group (P> 0.05). The serum IgE levels of all genotypes in the control group were lower than those in the asthma group (P <0.05) . Conclusion The rs4129267 polymorphism of IL-6R gene is associated with the susceptibility to asthma in Han children, but not to the level of serum IgE in children with asthma.
