目的:探讨胰岛素样生长因子-1(Insulin-like-growth factor-1,IGF-1)在体外对胶质瘤C6细胞垂体瘤转化基因(pituitary tumor transforming gene,PTTG)表达的影响。方法:体外培养胶质瘤C6细胞,给与不同浓度的IGF-1(0.1、1.0、10.0ng/ml)作用24h后半定量逆转录聚合酶链式反应(RT-PCR)法检测PTTG mRNA,Western blot检测PTTG蛋白表达。结果:不同浓度IGF-1作用后C6细胞PTTGmRNA表达与蛋白水平均增高,差异有统计学意义(P<0.01),且随着浓度的增加,其升高越明显。结论:IGF-1可以上调PTTG的表达,并剂量依赖性。
Objective: To investigate the effect of Insulin-like-growth factor-1 (IGF-1) on the expression of pituitary tumor transforming gene (PTTG) in glioma C6 cells in vitro. Methods: The C6 glioma cells were cultured in vitro and the expression of PTTG mRNA was detected by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) after treated with different concentrations of IGF-1 (0.1,1.0,10.0ng / ml) Western blot detection of PTTG protein expression. Results: The expression of PTTG mRNA and the protein level of C6 cells increased with the different concentrations of IGF-1, the difference was statistically significant (P <0.01), and with the increase of concentration, the more obvious the increase. Conclusion: IGF-1 can up-regulate PTTG expression in a dose-dependent manner.
