优降糖是有代表性的第二代磺脲药,为了研究它在体内的代谢变化,我们建立了血浆优降糖浓度高压液相色谱测定法(HPLC法),并进行了正常人口服优降糖的药代动力学研究。测定血浆优降糖HPLC法:内标法定量测定,座标系呈高度直线相关,Y=0.99999,线性范围25~400ng/ml。回归方程:X=0.3918Y~0.0017(X=浓度比,Y=相应校正因子)。高、中、低浓度的优降糖标准血浆样品的批间、批内变异系数均<11%。平均回收率:高浓度=101.4±7.6%,中浓度=103.3±11.4%,低浓度=98.8±8.0%。
