目的优选骨伤凝胶膏剂的促渗剂,研究其透皮吸收行为和体外释放度。方法采用改良Franzs扩散池法,生理盐水为接受液,用高效液相色谱法测定,以柚皮苷24 h累积透过量和增渗倍数为指标,优选骨伤凝胶膏剂的促渗剂并测定其释放度和透皮吸收情况。结果选用氮酮和丙二醇为促渗剂时,透皮吸收较好,最后确定氮酮和丙二醇的比例为(1∶1),用量为5%;柚皮苷24 h累积释放率为83.48%,累积透皮吸收率为46.59%,体外释药和透皮吸收均符合威布尔分布方程。结论骨伤凝胶膏剂加入氮酮和丙二醇后,透皮吸收明显增加,透皮吸收和释药均良好。
OBJECTIVE: To optimize the penetration enhancer of bone-wound gel plaster and study its transdermal absorption and in vitro release. Methods The modified Franzs diffusion cell method and physiological saline were used as the receiving solution. The content of translucency and infiltration multiplication of naringin at 24 h were determined by high performance liquid chromatography (HPLC) Its release and transdermal absorption. Results When using Azone and Propylene Glycol as penetration enhancers, transdermal absorption was better. Finally, the ratio of Azone and Propylene Glycol was determined to be 1: 1 with the dosage of 5%. The accumulative release rate of Naringin at 24 h was 83.48% The cumulative transdermal absorption rate was 46.59%. The in vitro drug release and transdermal absorption were in line with the Weibull distribution equation. Conclusion Osteogenetic gel ointment added azone and propylene glycol, transdermal absorption increased significantly transdermal absorption and release were good.
