目的:探究贮藏温度对不同含水量白术种子质量的影响,为其贮藏提供理论依据。方法:利用硅胶干燥法获得9.24%、7.89%、5.50%、4.30%四个含水量的白术种子,以未干燥的种子(含水量12.05%)为对照,在4℃和室温下贮藏1年后,测定种子发芽率、抗氧化酶活性及可溶性蛋白质含量等指标。结果:随着种子不断脱水,发芽率、生活力降低,浸出液电导率升高,MDA先升高后降低,可溶性蛋白质总体降低,SOD、POD酶活性随含水量降低总体升高。贮藏1年后,4℃下不同含水量的种子发芽率都高于68.89%,抗氧化酶活性高,而在室温下贮藏的种子劣变程度较明显,除含水量为5.50%的种子发芽率能够保持在40.00%外,其余处理均低于29.00%,且抗氧化酶活性较低。5.50%含水量的种子在不同温度下贮藏1年后,种子的发芽率、生活力和抗氧化酶活性均高于其他处理,电导率和MDA含量均较低。结论:白术种子具有耐脱水特性,在4℃下有较好的贮藏效果,属于正常性种子。将5.50%确定为本次试验所得5个含水量中白术种子的最适贮藏含水量。
Objective: To investigate the influence of storage temperature on seed quality of Atractylodes macrocephala with different water content, and to provide a theoretical basis for its storage. Methods: Four kinds of Atractylodes macrophyllus seeds with water content of 9.24%, 7.89%, 5.50% and 4.30% were obtained by silica gel drying method. The dried seeds of Atractylodes macrocephala (moisture content 12.05%) were used as control and stored at 4 ℃ for 1 year The seed germination rate, antioxidant enzyme activity and soluble protein content were measured. Results: With the continuous dehydration of seeds, the germination rate and viability decreased, and the conductivity of leachate increased. MDA increased first and then decreased, soluble protein decreased, and SOD and POD activities increased with the decrease of water content. After 1 year of storage, the germination percentage of seeds with different water contents at 4 ℃ was higher than 68.89%, and the activity of antioxidant enzymes was high. However, the seed deterioration degree of stored seeds at room temperature was more obvious. Except the seed germination rate of 5.50% Can keep at 40.00%, other treatments are less than 29.00%, and the antioxidant enzyme activity is low. Seed germination rate, viability and antioxidant enzyme activity of seeds with 5.50% water content after being stored for 1 year at different temperatures were higher than those of other treatments, and their conductivity and MDA content were lower. Conclusion: Atractylodes macrocephala has dehydration resistance and good storage effect at 4 ℃, which belongs to normal seed. The 5.50% is determined as the optimum moisture content of the seed of Atractylodes macrocephala in the five water content obtained in this experiment.
