目的比较乳腺癌组织和癌旁正常组织中miRNA-192的表达是否有差异及其与病理类型和病变进展之间的关系。方法通过整群抽样的方法选取19例乳腺癌患者手术切除的乳腺癌组织和癌旁正常乳腺组织,采用TRIzol法提取标本组织的总RNA,利用SYBR Green实时荧光定量PCR方法对miRNA-192进行定量检测,比较乳腺癌组织和癌旁正常乳腺组织miRNA-192的表达是否有差异。结果 miRNA-192在乳腺癌组织中表达量(ΔCt值)为10.58±3.78,在正常乳腺组织的表达量(ΔCt值)为11.16±3.40,差异无统计学意义。经Spearman相关分析,乳腺癌组织中的miRNA-192表达倍数(2-ΔΔCt值)与患者年龄、肿瘤大小、淋巴结转移、妊娠次数、肿瘤分期、生产次数、孕激素分泌无相关关系,仅与雌激素分泌呈负相关(P<0.01);经逐步法线性回归分析,以miRNA-192表达倍数为因变量时,各临床资料变量均未进入回归方程。结论乳腺癌患者癌组织及癌旁正常乳腺组织中miRNA-192的表达无明显差别。
Objective To compare the differences of miRNA-192 expression in breast cancer tissues and adjacent normal tissues and its relationship with pathological types and pathological changes. Methods Totally 19 breast cancer tissues and normal breast tissues were selected by cluster sampling method. Total RNA was extracted by TRIzol method and quantified by SYBR Green real-time PCR Detect and compare the expression of miRNA-192 in breast cancer tissues and adjacent normal breast tissues. Results The expression of miRNA-192 in breast cancer tissues (ΔCt value) was 10.58 ± 3.78, and in normal breast tissues (ΔCt value) was 11.16 ± 3.40, the difference was not statistically significant. According to Spearman correlation analysis, the expression of miRNA-192 (2-ΔΔCt value) in breast cancer tissues had no correlation with patient age, tumor size, lymph node metastasis, number of pregnancy, tumor staging, production frequency and progesterone secretion, Hormone secretion was negatively correlated (P <0.01). After stepwise linear regression analysis, when the miRNA-192 expression multiple was taken as the dependent variable, all the clinical data variables did not enter the regression equation. Conclusion There is no significant difference in the expression of miRNA-192 between breast cancer tissues and adjacent normal breast tissues.
