目的:研究氢化可的松对人表皮共培养体系(黑素细胞与角质形成细胞)酪氨酸酶活性及黑素生成的影响。方法培养人表皮黑素细胞和人表皮黑素细胞与角质形成细胞共培养体系,利用四甲基偶氮唑蓝(MTT)还原法测定细胞活力,测定酪氨酸酶活性,475nm 比色法测定黑素含量。结果10-7mol/L、10-6μmol/L和10-5mol/L氢化可的松对于黑素细胞及共培养细胞酪氨酸酶活性及黑素生成均有较强的剂量相关性增强作用,10-6μmol/L和10-5mol/L氢化可的松与对照组相比有显著差异(p<0.01)。结论氢化可的松对于共培养体系酪氨酸酶活性及黑素生成有较强的剂量相关性增强作用。“,”Objective To study the effects of hydrocrtison on the tyrosinase activity and melanogenesis in co-culture model of human melanocytes and keratinocytes.Methods The model of co-culture of human melanocytes and keratinocytes was cultured and MTT was used to measure the proliferation of melanocytes and the co-cultures.The tyrosinase activity and melanin con-tent were measured.Results The melanin synthesis and tyrosinase activity were clearly enhanced by hydrocrtison in a dose-dependent manner at concentrations of 10 -7mol/L、10 -6μmol/Land 10-5mol/L in both melanocytes and co-cultures.10 -6μmol/Land 10 -5mol/L of hydrocrtison showed statistical y significant suppression (p <0.01)than the control.Conclusion Hydrocrtison exhibits a dose-dependent enhancing ef ects on melanin synthesis and tyrosinase activity in co-cultures of human melanocytes and keratinocytes.
