目的:采用三磷酸腺苷法(ATP-TCA)对口腔颊黏膜鳞癌组织标本进行肿瘤药敏检测,为口腔颊黏膜鳞癌的个体化化疗选择提供依据。方法:45例口腔颊黏膜鳞癌新鲜标本行肿瘤细胞分离,6种化疗药物分别和癌细胞原代培养后,进行ATP-TCA检测。结果:所有标本均顺利完成检测,可评估率为100%,单药强敏感率为吡柔比星44.44%、替尼泊苷35.56%、表柔比星31.11%、顺铂22.22%、氟尿嘧啶20.00%、紫杉醇13.33%,不同药物对口腔颊黏膜鳞癌的强敏感程度有差异(P=0.012),该组不同个体肿瘤细胞对不同药物的敏感性程度差异具有显著统计学意义(P=0.0006)。结论:ATP-TCA可为口腔颊黏膜鳞癌的临床个体化化疗提供重要的指导作用。
OBJECTIVE: To detect the drug susceptibility of buccal mucosal squamous cell carcinoma tissues by adenosine triphosphate (ATP-TCA) assay and to provide evidence for the selection of personalized chemotherapy for buccal mucosal squamous cell carcinoma. Methods: 45 cases of oral buccal mucosa squamous cell carcinoma were isolated from tumor cells. Six kinds of chemotherapeutic drugs and primary cultured cancer cells were used to detect ATP-TCA. Results: All the samples were successfully tested. The rate of single drug sensitivity was 44.44% for pirarubicin, 35.56% for teniposide, 31.11% for epirubicin, 22.22% for cisplatin and 20.00 for fluorouracil %, And paclitaxel 13.33%. The sensitivity of different drugs to oral buccal mucosal squamous cell carcinoma was significantly different (P = 0.012). The sensitivity of different individual tumor cells to different drugs was significantly different (P = 0.0006) . Conclusion: ATP-TCA can provide important guidance for personalized chemotherapy of oral buccal squamous cell carcinoma.
