脐血作为造血于/祖细胞的来源具有诸多优点,应用前景广泛.采用多种造血生长因子组合,建立体外大量扩增脐血于/祖细胞的培养体系,已逐渐成为人们关注的热点.本研究动态观察了脐血单个核细胞(MNC)经不同造血生长因子组合刺激的增殖效应.结果显示,①脐血MNC培养至11~20d时,达到增殖高峰,21d后细胞增殖明显变缓.20d时,不同的造血生长因子组合对脐血造血于/祖细胞扩增倍数不同,结果如下,(a)rhIL-1+rhIL-3+rhIL-6+rhG-CSF+rhGM-CSF+rhSCF组合使有核细胞总数增殖为初始的43倍;
