目的鉴定细胞凋亡的一个重要生化指标是凋亡细胞的核 DNA通过琼脂糖凝胶电泳呈现典现的 DNA“梯状”图谱。传统分离凋亡细胞片段化 DNA的方法多采用酚 /氯仿抽提。因此 ,有必要探寻一种更为灵敏、简单的分离凋亡片段化 DNA的新方法。方法通过比较传统分离凋亡片段化 DNA的方法 ,并对 HERRMANN等 [1 ]报道的方法加以改进。结果本文介绍一种快速分离凋亡 DNA片段化的新方法 ( NP- 40法 )。结论这种简单、快速的分离方法无需酚 /氯仿反复抽提 ,无需大量的细胞样品 ,并且凋亡 DNA片段化梯状图谱清晰 ,适用于大多数凋亡细胞片段化 DNA的分离
Aim To identify an important biochemical indicator of apoptosis as a DNA “ladder” map of the apoptotic cell nuclear DNA present on agarose gel electrophoresis. Traditional methods for the isolation of apoptotic DNA fragments by phenol / chloroform extraction. Therefore, it is necessary to explore a more sensitive and simple method for the isolation of apoptotic fragmented DNA. Methods By comparing the traditional methods for the isolation of apoptotic fragmented DNA, the methods reported by HERRMANN et al. [1] were improved. Results In this paper, a novel method for the rapid isolation of apoptotic DNA fragmentation (NP-40 method) was introduced. Conclusions This simple, rapid separation method eliminates the need for repeated extraction with phenol / chloroform, eliminates the need for extensive cell samples, and provides a clear, step-by-step ladder of apoptotic DNA that is suitable for the isolation of most apoptotic cell fragmented DNA
