以中牟 5号大蒜根尖为外植体 ,在 1 1种固体培养基上诱导愈伤组织 ,筛选出最佳培养基MS + 2 ,4-D 0 .5mg/L和MS + 2 ,4-D 2 .0mg/L +KT 0 .5mg/L。愈伤组织形成后在上述1 1种培养基上继代 3~ 4次 ,获得胚性愈伤组织后再转至液体培养基中 ,经强化培养 3个月后获得胚性细胞悬浮系。试验结果表明 :缩短换液时间 (每隔 3~ 4天换液 1次 ) ,可加快胚性细胞悬浮系的建立
The root tips of Zhongmou No.5 garlic were used as explants to induce the callus on 11 kinds of solid medium and the best medium MS + 2, 4-D 0.5mg / L and MS + 2, 4- D 2 .0 mg / L + KT 0 .5 mg / L. After the formation of callus in the above 1 1 kinds of medium subculture 3 to 4 times, embryogenic callus obtained after transfer to liquid medium, after 3 months of intensive culture obtained embryogenic cell suspension system.
