目的通过观察补中益气汤对肺腺癌荷瘤裸鼠移植瘤中LRP蛋白表达影响,探讨补中益气汤治疗肺腺癌的作用机制。方法体外培养A549/DDP及A549细胞,接种于BALB/c nu/nu裸鼠腋窝皮下,荷瘤成功后给予低、中、高剂量补中益气汤治疗。实验共分为10组:即A组(A549/DDP荷瘤对照组)、B组(A549/DDP荷瘤+顺铂组)、C组(A549/DDP荷瘤+顺铂+低剂量补中益气汤组)、D组(A549/DDP荷瘤+顺铂+中剂量补中益气汤组)、E组(A549/DDP荷瘤+顺铂+高剂量补中益气汤组)、a组(A549荷瘤对照组)、b组(A549荷瘤+顺铂组)、c组(A549荷瘤+顺铂+低剂量补中益气汤组)、d组(A549荷瘤+顺铂+中剂量补中益气汤组)、e组(A549荷瘤+顺铂+高剂量补中益气汤组)。测量移植瘤体积并计算药物抑瘤率,蛋白质印迹法检测移植瘤组织LRP蛋白的表达。结果顺铂对A549/DDP移植瘤体积无显著影响(P>0.05),而使A549移植瘤体积显著缩小(P<0.05);补中益气汤使A549/DDP及A549移植瘤体积均缩小(P<0.05,P<0.05),顺铂抑瘤率均增加(P<0.05,P<0.05),随着中药剂量的增加,移植瘤体积逐渐缩小,顺铂抑瘤率逐渐增加;顺铂使A549/DDP移植瘤组织中LRP蛋白表达增加(P<0.05),A549移植瘤组织中LRP蛋白表达减少(P<0.05);补中益气汤使A549/DDP移植瘤组织中LRP蛋白表达减少(P<0.05),以中剂量效果最为显著,补中益气汤使A549移植瘤组织中LRP蛋白表达也减少(P<0.05),但低、中、高剂量之间效果无显著差异。结论在对顺铂敏感及顺铂耐药的肺腺癌移植瘤组织中,补中益气汤均可使移植瘤体积缩小,顺铂的抑瘤率增加,LRP蛋白的表达下调。
Objective To observe the effect of Buzhong Yiqi Decoction on the expression of LRP protein in lung adenocarcinoma xenografts in nude mice and to explore the mechanism of Buzhong Yiqi Decoction in treating lung adenocarcinoma. Methods A549 / DDP and A549 cells were cultured in vitro and inoculated subcutaneously into the armpit of BALB / c nu / nu nude mice. After the tumor was successfully treated, low, medium and high dose Buzhong Yiqi Decoction was given. The experiment was divided into 10 groups: group A (A549 / DDP tumor bearing control group), group B (A549 / DDP tumor bearing + cisplatin group), group C (A549 / DDP tumor bearing + cisplatin + D group (A549 / DDP bearing tumor + cisplatin + middle dose Bu Zhong Yi Qi Tang group), E group (A549 / DDP bearing tumor + cisplatin + high dose Bu Zhong Yi Qi Tang group) Group A (A549 tumor-bearing control group), Group b (A549 tumor + cisplatin group), Group C (A549 tumor + cisplatin + low dose Buzhong Yiqi Decoction group), group d Platinum + dose Bu Zhong Yi Qi Tang group), e group (A549 tumor + cisplatin + high-dose Bu Zhong Yi Qi Tang group). The volume of xenograft tumor was measured and the rate of tumor inhibition was calculated. The expression of LRP protein in xenograft tumor was detected by Western blotting. Results The cisplatin had no significant effect on the volume of A549 / DDP xenografts (P> 0.05), and the volume of A549 xenografts was significantly reduced (P <0.05). The volume of A549 / DDP and A549 xenografts was reduced by Buzhong Yiqi Decoction (P <0.05, P <0.05). The inhibitory rate of Cisplatin was also increased (P <0.05, P <0.05). With the increase of the dose of traditional Chinese medicine, the volume of xenograft tumor gradually decreased and the inhibitory rate of cisplatin increased gradually. The expression of LRP protein in A549 / DDP xenografts was increased (P <0.05), and the expression of LRP protein in A549 xenografts was decreased (P <0.05). Buzhong Yiqi decoction reduced the expression of LRP protein in A549 / DDP xenografts (P <0.05). The effect of Zhongzhongyiqi Decoction was the most significant. The expression of LRP protein in A549 xenografts was also decreased (P <0.05), but there was no significant difference between low, middle and high doses. CONCLUSION: Buzhongyiqi Decoction can reduce the volume of xenograft tumor, increase the inhibitory rate of cisplatin, and down-regulate the expression of LRP protein in cisplatin-sensitive and cisplatin-resistant lung adenocarcinoma xenografts.
