[目的]研究独活中二氢欧山芹醇-β-D葡萄糖苷和二氢欧山芹醇的最佳提取条件,同时建立了同时测定独活中二氢欧山芹醇-β-D葡萄糖苷和二氢欧山芹醇含量的高效液相色谱法(HPLC)。[方法]通过单因素分析法考察了提取溶剂、料液比、超声提取时间对独活提取物中二氢欧山芹醇-β-D-葡萄糖苷和二氢欧山芹醇含量的影响。采用HPLC法对不同提取条件和不同产地的独活中两种成分含量。[结果]独活中二氢欧山芹醇-β-D葡萄糖苷和二氢欧山芹醇的最佳提取条件为提取溶剂用100%的甲醇,料液比为1∶50,超声提取60min。二氢欧山芹醇-β-D-葡萄糖苷在0.3125~20μg/m L内呈良好线性,平均回收率为99.0%(RSD=2.99%);二氢欧山芹醇在0.3125~20μg/m L内呈良好线性,平均回收率为102%(RSD=2.59%)。[结论]所建立的高效液相色谱法,能够准确测定独活中二氢欧山芹醇-β-D葡萄糖苷和二氢欧山芹醇含量,可用于独活药材的质量评价。
[Objective] The study aimed to study the optimal extraction conditions of dihydroalpinechol-β-D-glucoside and dihydro-parsnant alcohol in the independent living, and establish the simultaneous determination of dihydro-isooctyl alcohol-β-D glucoside and dihydropalmatine High Performance Liquid Chromatography (HPLC). [Method] The effects of extraction solvent, solid-to-liquid ratio and ultrasonic extraction time on the content of dihydropalmatin-β-D-glucoside and dihydropalphane in the extract of Ligustici mongolicum were investigated by single factor analysis. Using HPLC method for different extraction conditions and independent living in the two components of the content. [Result] The optimum extraction conditions of dihydropalmatin-β-D-glucoside and dihydrocapspinal alcohol were as follows: extraction solvent was 100% methanol, the ratio of material to liquid was 1:50, ultrasonic extraction for 60min. Dihydropalmatine alcohol-β-D-glucoside showed a good linearity in the range of 0.3125 ~ 20μg / m L with an average recovery of 99.0% (RSD = 2.99%). Dihydrocapsalin was good at 0.3125 ~ 20μg / mL Linear, with an average recovery of 102% (RSD = 2.59%). [Conclusion] The established HPLC method can accurately determine the content of dihydrocapspinal alcohol-β-D-glucoside and dihydropalphane in the independent living, and can be used for the quality evaluation of the independent living medicinal materials.
