目的 探讨 VEGF高表达的卵巢癌中 VEGF基因结构有无异常 .方法 用 RT- PCR方法自胎盘中克隆 VEGFc DNA以作为杂交探针 ;收集临床卵巢癌手术切除标本 ,以正常卵巢组织为对照 ,从中提取基因组 DNA并进行 Southern印迹杂交 .结果 序列分析表明所获得基因正确 ,Southernblot分析 10 / 19呈现出大于 2 3 kb片段和 4.3 kb片段的异常片段 ,而不同于正常对照组织的大于 30 kb,6 .5 kb和 2 .5 kb.结论 VEGF高表达的卵巢癌中有该基因的重排
Objective To investigate the VEGF gene structure in ovarian cancer with overexpression of VEGF.Methods VEGFc DNA was cloned from the placenta using RT-PCR as a hybridization probe.Clinical specimens of ovarian cancer were collected and compared with normal ovarian tissue Genomic DNA was extracted and Southern blot hybridization was performed.Results Sequence analysis showed that the obtained gene was correct, and Southern blot analysis showed that abnormal fragments larger than 23 kb and 4.3 kb were found in 10/19 Southern blot analysis, which was different from the normal control .5 kb and 2.5 kb.
