目的:HPLC法同时测定乌头属药材附子、川乌、草乌、雪上一枝蒿中乌头碱(AC)、次乌头碱(HA)、新乌头碱(MA)、苯甲酰新乌头原碱(BMC)的含量。方法:色谱柱:Phenomenex-ODS(250 mm×4.0 mm,5μm);流动相:乙腈-四氢呋喃(25∶15),梯度洗脱,流速:1.0 mL/min;检测波长:235 nm;柱温:30℃。结果:4种乌头类生物碱的线性范围分别为:0.0241~0.5211μg(r1=0.9995)、0.0168~0.3152μg(r2=0.9994)、0.0107~0.2674μg(r3=0.9994)、0.0122~0.2503μg(r4=0.9996);4种药材中4种生物碱的加样回收率范围分别为:附子:95.56~98.63%、川乌:98.83~100.47%、草乌:95.91~99.30%、雪上一枝蒿:95.99%~98.53%,RSD%≤4.0%。结论:乌头属4种药材4种生物碱含量差异较大,为临床疾病治疗防止中毒反应的发生提供了一定的参考依据。
Objective: To determine the contents of aconitine (AC), hypaconitine (HA), mesaconitine (MA), benzo [ Head protamine (BMC) content. The mobile phase consisted of acetonitrile-tetrahydrofuran (25:15), gradient elution, the flow rate was 1.0 mL / min, the detection wavelength was 235 nm, the column temperature was 250 ℃, 30 ° C. Results: The linear ranges of four aconitines were 0.0241 ~ 0.5211μg (r1 = 0.9995), 0.0168 ~ 0.3152μg (r2 = 0.9994), 0.0107 ~ 0.2674μg (r3 = 0.9994), 0.0122 ~ 0.2503μg r4 = 0.9996). The range of recoveries of four alkaloids in four herbs were as follows: Aconite: 95.56 ~ 98.63%, Chuanwu: 98.83 ~ 100.47%, Aconitum: 95.91 ~ 99.30%, Artemisia selengensis: 95.99 % ~ 98.53%, RSD% ≤4.0%. Conclusion: The content of four kinds of alkaloids in four kinds of medicinal materials of Aconitum are quite different, which provides a reference for the prevention of toxic reactions in the treatment of clinical diseases.
