目的:优选酶解法辅助提取钩藤中钩藤碱的工艺条件,为该药材的资源利用提供参考。方法:采用HPLC测定钩藤碱含量,流动相甲醇-水(60∶40),检测波长254 nm。以钩藤碱提取率为指标,在单因素试验基础上,通过正交试验考察加酶量、酶解温度、酶解时间、酶解p H对酶解提取工艺的影响。结果:最佳酶解提取工艺条件为纤维素酶用量1%,p H 4.5,酶解温度50℃,酶解时间2 h。钩藤碱提取率0.54%,较加热回流法提高了54.29%。结论:酶解法有利于提高钩藤碱的得率,优选的提取工艺稳定可行,适合推广于药材中生物碱类成分的提取。
OBJECTIVE: To optimize the technological conditions for the extraction of rhynchophylline from Uncaria rhynchophylla by enzymatic hydrolysis, and to provide a reference for the resource utilization of this herb. Methods: The content of rhynchophylline was determined by HPLC. The mobile phase consisted of methanol - water (60:40) and the detection wavelength was 254 nm. Taking rhynchophylline extraction rate as an index, on the basis of single factor test, the effect of adding enzyme amount, enzymolysis temperature, enzymolysis time and enzymatic hydrolysis pH on the enzymolysis extraction process was investigated by orthogonal test. Results: The optimal conditions of enzymatic extraction were 1% cellulase, p H 4.5, enzymolysis temperature 50 ℃, enzymolysis time 2 h. Rhynchophylline extraction rate of 0.54%, compared with heating reflux method increased 54.29%. Conclusion: Enzymolysis is helpful to improve the yield of rhynchophylline. The optimal extraction process is stable and feasible. It is suitable for the promotion of the extraction of alkaloids from medicinal materials.
