目的:研究与子宫内膜癌淋巴转移相关的蛋白质。方法:提取正常子宫内膜、淋巴结阴性和淋巴结阳性的子宫内膜癌组织总蛋白,用双向凝胶电泳分离蛋白质,用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)分析鉴定差异蛋白点。结果:在淋巴结阴性和淋巴结阳性的子宫内膜癌中,共筛选出11个差异蛋白,与淋巴结阴性组比较,淋巴结阳性内膜癌中,热休克蛋白27、3种肌动蛋白、S100钙结合蛋白A9、丙酮酸激酶、α烯醇化酶、白蛋白和组氨酸三聚体核苷结合蛋白1表达上调,天冬酰胺酶样1蛋白和酯酶D/甲酰-谷胱甘肽水解酶下调。结论:淋巴结阳性和阴性子宫内膜癌的双向凝胶电泳图谱之间存在较明显差异。经鉴定的9种蛋白质可能在子宫内膜癌发生淋巴转移过程中发挥作用。
Objective: To study the proteins related to lymphatic metastasis of endometrial cancer. Methods: The total protein of normal endometrial, lymph node-negative and lymph node-positive endometrial cancer tissues was extracted and the proteins were separated by two-dimensional gel electrophoresis. The differential proteins were identified by matrix-assisted laser desorption / ionization time of flight mass spectrometry (MALDI-TOF-MS) point. Results: Eleven differential proteins were screened in endometrial cancer with negative lymph node and lymph node, and compared with lymph node negative group, there were 27 heat-shock protein, actin, S100 calcium in lymph node positive endometrial carcinoma Protein A9, pyruvate kinase, alpha-enolase, albumin and histidine trimer nucleoside binding protein 1 are upregulated in expression, asparaginase-like 1 protein and esterase D / formyl-glutathione hydrolase Down. Conclusion: Two-dimensional gel electrophoresis patterns of lymph node-positive and -negative endometrial carcinoma have obvious differences. The identified nine proteins may play a role in the lymphatic metastasis of endometrial carcinoma.
