作者用鼠淋巴细胞杂交瘤技术,将自身免疫MRL/lpr/lpr小鼠的脾细胞与非分泌性骨髓瘤(P3X63-Ag8653)在PEG作用下融合,获得了一种单克隆抗DNA自身抗体(EM85).经双向免疫扩散试验证明,该抗体是一种IgG2a抗体.用多种核酸和合成的多核苷酸以纤维素脂滤膜放射免疫试验,通过竞争性实验分析了这种单克隆抗体(mAb)的特异性,发现用ss-DNA、ds-DNA、poly(dC)、poly(dG)、poly(G)、poly(I)和poly(dA)、poly(dT)时可见明显的抑制,指示EM85 mAb识别ds-DNA、ss-DNA和多种多核苷
