目的比较黄芪、水蛭及其配方含药血清对大鼠系膜细胞生长周期、凋亡的影响。方法制备水蛭高浓度(水蛭组)、黄芪高浓度(黄芪组)和水蛭高黄芪低浓度(配方组)含药血清,并对大鼠系膜细胞(MC)进行干预培养,应用流式细胞仪检测MC生长周期与凋亡,显微镜观察凋亡细胞的形态。结果各组含药血清均能使大量MC处于G0/G1期,与对照组比较差异有统计学意义(P<0.01);各组亦均能提高大鼠MC凋亡率,水蛭组、配方组与对照组比较差异有统计学意义(P<0.01)。结论水蛭、黄芪及其配方含药血清均能阻止MC进入S期从而达到抑制增生的目的,且能提高MC的凋亡率。
Objective To compare the effects of astragalus membranaceus, leeches and their formula-containing serum on the growth cycle and apoptosis of rat mesangial cells. Methods The serum containing the high concentration of leech (water blister group), the high concentration of jaundice (Huangqi group) and the high concentration of jaundice (formula group) were prepared, and the rat mesangial cells (MC) were cultured, and the flow cytometer was used. The growth cycle and apoptosis of MC were detected and the morphology of apoptotic cells was observed under a microscope. Results All groups of drug-containing serum could make a large number of MCs in G0/G1 phase, and the difference was statistically significant compared with the control group (P<0.01). Each group also increased the apoptosis rate of MC in rats. Compared with the control group, the difference was statistically significant (P<0.01). Conclusion Both leeches, astragalus membranaceus and its formula-containing serum can prevent MC from entering S phase and achieve the purpose of inhibiting proliferation, and can increase the apoptosis rate of MC.
