[目的]探明α-倒捻子素对芒果炭疽病菌麦角甾醇合成的影响。[方法]通过测定α-倒捻子素对芒果炭疽病菌菌丝生长毒力,建立超高效液相色谱法(UPLC)分析其对菌丝麦角甾醇含量的影响。[结果]采用UPLC法检测,麦角甾醇的保留时间为2.58 min,在2.5~100 mg/L时线性相关系数为0.9999,平均回收率为85.72%,相对标准偏差为4.24%。α-倒捻子素对芒果炭疽病菌菌丝生长的EC_(50)和EC_(80)值分别为73.01、403.96 mg/L;在生长抑制剂量下,α-倒捻子素显著降低病原菌中麦角甾醇含量(P<0.05),且对麦角甾醇合成抑制作用呈现剂量依赖性。[结论]UPLC法适用于麦角甾醇的快速检测,α-倒捻子素对芒果炭疽病菌生长和麦角甾醇合成具有明显抑制作用。
[Objective] The research aimed to investigate the effect of α-mangostin on the ergosterol synthesis of anthracnose in mango. [Method] The effects of α-mangostin on the mycelial growth of the mycelium of mango anthracnose were studied by UPLC. The effects of α-mangostin on the mycelial ergosterol content of mycelia were analyzed by UPLC. [Result] The retention time of ergosterol by UPLC method was 2.58 min. The linear correlation coefficient at 2.5-100 mg / L was 0.9999 with the average recovery of 85.72% and the relative standard deviation of 4.24%. The EC 50 and EC 80 values of α-mangostin on mycelial growth of C. mango were 73.01 and 403.96 mg / L, respectively. In the growth inhibition dose, α-mangostin significantly reduced the content of ergosterol (P <0.05), and inhibited the synthesis of ergosterol in a dose-dependent manner. [Conclusion] The UPLC method is suitable for the rapid detection of ergosterol, and α-mangostin significantly inhibited the growth of C. mango and the synthesis of ergosterol.
