用PEG融合法融合甘薯品种高系14号(6x)及其近缘野生种Ipomoeatriloba(2x)的原生质体,融合率达28%。将融合原生质体培养在含有0.05mg·L-12,4-D和0.5mg·L-1激动素(KT)的改良MS培养基中,4~5d后,再生细胞发生首次细胞分裂。培养12周后,将直径约2.0mm的小愈伤组织转移到添加0.05mg·L-12,4-D的MS培养基上培养4周,愈伤组织迅速增殖。将其中的37个愈伤组织转移到添加0.2mg·L-1IAA和1.0mg·L-1BAP的MS培养基上,2个愈伤组织再生植株。将未再生植株的35个愈伤组织培养在MS基本培养基上,17个愈伤组织再生植株。由其中仅存的7个愈伤组织所获得的再生植株的染色体数为44~50,在形态学上相似于I.triloba,但又有一定变异,表明为体细胞杂种植株。
Protoplast fusion of sweet potato variety GaoXuan No.14 (6x) and its related wild species Ipomoeatriloba (2x) was achieved by PEG fusion method, and the fusion rate was 28%. The fusion protoplasts were cultured in modified MS medium containing 0.05 mg · L-12, 4-D and 0.5 mg · L -1 kinetin (KT). After 4 ~ 5 days, the first division of regenerated cells occurred. After 12 weeks of culture, small callus with diameter of about 2.0 mm was transferred to MS medium supplemented with 0.05 mg · L-12,4-D for 4 weeks, and the callus rapidly proliferated. Thirty-seven of them were transferred to MS medium supplemented with 0.2 mg · L-1 IAA and 1.0 mg · L-1BAP, and two callus were regenerated. Thirty-five callus from non-regenerated plants were cultured on MS basal medium and 17 callus plants were regenerated. The regenerated plants obtained from the remaining seven callus had chromosome numbers of 44-50, which are similar in morphology. Triloba, but with some variation, indicates somatic hybrid plants.
