目的:老鼠大脑组织所含成分和结构特殊,其组织与玻片粘附性差,在免疫组化染色和原位杂交实验中脱片率很高,故应用一种新型粘片剂APES(氨醛三己氧基硅烷)制作防脱片。方法:载玻片经酸浸泡、清洗、烤干、浸入1∶50丙酮稀释的APES中,浸泡20 s,再放入100%丙酮反复3次浸泡清洗,制作防脱片。结果:APES制作防脱片应用在免疫组化和原位杂交实验中防脱片效果稳定、可靠。结论:APES制作防脱片效果特好,方法简单,脑组织及其他组织实验都可应用,可在临床和科研实验室中推广使用。
OBJECTIVE: The composition and structure of rat brain tissue are special, its adhesion to tissue and slide is poor, and the rate of stripping is very high in immunohistochemical staining and in situ hybridization experiments. Methods: Slides were soaked in acid, washed, dried and soaked in 1:50 acetone-diluted APES for 20 seconds, then immersed in 100% acetone for 3 times to prepare anti-shedding tablets. Results: APES anti-shedding application in the immunohistochemistry and in situ hybridization experiments anti-shedding effect is stable and reliable. Conclusion: APES is a good anti-desquamating patch with a simple method and can be applied in both brain and other tissues. It can be used in clinical and research laboratories.
