目的:研究人类端粒酶逆转录酶(hTERT)基因的反义核酸(AS PS-ODN)能否增强复发急性淋巴细胞白血病原代细胞对顺铂的敏感性。方法:采用免疫荧光标记法检测hTERT蛋白表达水平,以台盼蓝拒染法计数细胞数,通过细胞形态学观察及流式细胞仪细胞周期的分析来检测凋亡。结果:hTERT基因反义核酸能降低hTERT蛋白的表达。顺铂与hTERT基因反义核酸联合应用能明显抑制培养原代细胞的增殖,与单用顺铂组比较P<0.05。凋亡检测,用药48 h后,单用顺铂、顺铂联用反义核酸或正义核酸组,经Hoechst33258和PI双染法观察细胞均表现典型的细胞凋亡的形态学改变;顺铂联用反义核酸组,细胞的凋亡率明显高于单用顺铂组(P<0.05)。结论:hTERT基因反义核酸能增强复发急性淋巴细胞白血病原代细胞对顺铂的敏感性。
OBJECTIVE: To investigate whether antisense nucleic acid (AS PS-ODN) of human telomerase reverse transcriptase (hTERT) gene can enhance the sensitivity of cisplatin to the primary cells of recurrent acute lymphoblastic leukemia. Methods: The expression of hTERT protein was detected by immunofluorescence staining. The number of cells was counted by trypan blue exclusion method. The apoptosis was detected by morphological observation and flow cytometry analysis. Results: Antisense hTERT gene could decrease the expression of hTERT protein. The combination of cisplatin and hTERT gene antisense nucleic acid can significantly inhibit the proliferation of primary cultured cells, compared with cisplatin alone (P <0.05). Apoptosis was detected 48 h after treatment with cisplatin, cisplatin antisense or sense nucleic acid group, Hoechst33258 and PI double staining showed that the cells showed typical morphological changes of apoptosis; cisplatin With antisense nucleic acid group, the apoptosis rate of cells was significantly higher than that of cisplatin group (P <0.05). Conclusion: hTERT gene antisense oligonucleotide can enhance the sensitivity of cisplatin to the primary cells of recurrent acute lymphoblastic leukemia.
