目的:探讨巨噬细胞集落刺激因子(Macrophagecolony-stimulatingfactor,M-CSF)抗单核/巨噬细胞氧化损伤的作用机理,揭示M-CSF对小鼠巨噬细胞系RAW264.7细胞抗氧化系统影响。方法:以L929细胞条件培养液为M-CSF来源,采用酶活性测定等生化测定方法,考察M-CSF细胞内还原型谷胱甘肽含量、脂质过氧化物含量、过氧化氢酶活性、谷胜甘肽还原酶活性等检测指标的影响。结果:M-CSF处理可提高RAW264.7细胞内的过氧化氢酶活性及还原型谷胱甘肽含量,降低脂质过氧化物含量,且使细胞在tbOOH攻击下保持较高的抗氧化水平。结论:M-CSF可能通过改善单核巨噬细胞的氧化还原代谢而减轻细胞的过氧化损伤。
Objective: To investigate the mechanism of macrophage colony-stimulating factor (M-CSF) against oxidative damage of monocyte / macrophage and to reveal the effect of M-CSF on the anti-oxidative system of mouse macrophage cell line RAW264.7 . Methods: L929 cells conditioned medium was used as M-CSF source. The biochemical assay was used to determine the contents of reduced glutathione, lipid peroxidation and catalase in M-CSF cells. Glutathione reductase activity and other indicators of detection. Results: M-CSF treatment increased catalase activity and reduced glutathione content in RAW264.7 cells, decreased lipid peroxidation and maintained high levels of antioxidation under tbOOH challenge . Conclusion: M-CSF may reduce the cellular peroxidation injury by improving the redox metabolism of monocyte-macrophage.
